Overview

PAM37 is a Gs-biased positive allosteric modulator (PAM) of the beta-2 adrenergic receptor (beta2AR). It is being investigated for its potential to treat chronic asthma by mitigating airway remodeling and enhancing the bronchodilatory effects of beta-agonists. PAM37 specifically potentiates beta2AR-mediated cAMP-PKA signaling in airway smooth muscle (ASM) cells, leading to the inhibition of ASM proliferation induced by mitogens like PDGF or FBS. Research indicates that PAM37 prolongs and amplifies the signaling response of specific beta-agonists (such as isoproterenol, formoterol, and albuterol) but not others (like fenoterol or salmeterol), suggesting a conformation-specific binding mechanism that could be therapeutically advantageous in reducing disease severity and fixed airflow limitation.
