Overview

PAMA-co-PLAMA glycopolymer is an experimental cationic gene delivery nanosystem specifically designed for targeting hepatocellular carcinoma (HCC). It is a copolymer of 2-aminoethyl methacrylate hydrochloride (PAMA) and 2-lactobionamidoethyl methacrylate (PLAMA), synthesized using activators regenerated by electron transfer (ARGET) atom transfer radical polymerization (ATRP). The PLAMA component features lactobionic acid moieties that serve as ligands for the asialoglycoprotein receptor (ASGPR), which is highly expressed on HCC cells, enabling receptor-mediated endocytosis. This glycopolymer forms 'glycoplexes' with therapeutic DNA, such as the HSV-TK suicide gene, providing a biocompatible and efficient platform for targeted gene therapy. In preclinical studies, it has demonstrated high transfection efficiency and potent antitumor activity, particularly when combined with low doses of docetaxel.
