PAMAM-G3 is a third-generation polyamidoamine (PAMAM) cationic dendrimer being investigated for its therapeutic potential in oncology, particularly in inhibiting cancer metastasis. It functions as a molecular scavenger that utilizes its high positive surface charge to bind and neutralize negatively charged, pro-inflammatory, and pro-metastatic mediators, such as cell-free DNA (cfDNA) and tumor-derived extracellular vesicles (EVs). By sequestering these molecules, PAMAM-G3 prevents the activation of nucleic acid-sensing receptors, including Toll-like receptors (TLRs), which otherwise drive pro-invasive signaling pathways in both tumor cells and the host environment. Research primarily conducted at Duke University has demonstrated that PAMAM-G3 can significantly inhibit pancreatic cancer cell invasion in vitro and reduce liver metastasis in syngeneic murine models without affecting primary tumor growth.
