Overview

PAMAMG4-docosahexaenoic acid-paclitaxel conjugate is a dendrimer-based drug delivery system designed to improve the therapeutic index of paclitaxel in gastrointestinal cancers. It consists of a Generation 4 poly(amidoamine) (PAMAM) dendrimer conjugated to docosahexaenoic acid (DHA), an omega-3 fatty acid, and paclitaxel. The DHA component is intended to enhance cellular uptake and potentially reduce toxicity, while the dendrimer scaffold improves solubility and bioavailability. Preclinical studies in esophageal, gastric, and pancreatic cancer models have shown that this conjugate exhibits superior antiproliferative activity and induction of apoptosis (via P53 and P21 pathways) compared to free paclitaxel.
