Pamiparib is a potent and selective small molecule inhibitor of poly(ADP-ribose) polymerase 1 (PARP1) and PARP2. It is designed to exploit synthetic lethality in tumor cells with homologous recombination repair deficiencies, particularly those harboring BRCA mutations. By inhibiting PARP enzymes, pamiparib impairs the repair of single-strand DNA breaks, leading to accumulation of DNA damage and cell death in susceptible cancer cells. Pamiparib has demonstrated the ability to penetrate the blood-brain barrier and effectively trap PARP-DNA complexes. Developed by BeiGene, it is approved in China for treating patients with recurrent advanced ovarian cancer, fallopian tube cancer, or primary peritoneal cancer who have germline BRCA mutations and have received at least two prior lines of chemotherapy[1][3][5][7].
