Pamorelin is a sustained-release formulation of triptorelin embonate, a synthetic decapeptide agonist of the gonadotropin-releasing hormone (GnRH) receptor. It functions by initially stimulating the pituitary gland to release luteinizing hormone (LH) and follicle-stimulating hormone (FSH), followed by a period of receptor downregulation and desensitization. This results in a profound suppression of the pituitary-gonadal axis and a significant reduction in the production of sex steroids. In the context of clinical research led by Kenny Rodriguez-Wallberg (the ProFertil trial), Pamorelin is being evaluated in Phase 3 trials for its potential to protect ovarian function and preserve fertility in young women and teenagers undergoing gonadotoxic chemotherapy for various cancers, such as breast cancer, lymphoma, and leukemia. The induction of a temporary hypogonadotropic state is hypothesized to shield the ovaries from chemotherapy-induced damage, thereby preventing premature ovarian insufficiency.
