Overview

Pamrevlumab + gemcitabine + erlotinib is a combination therapy under investigation primarily for the treatment of pancreatic ductal adenocarcinoma. This combination targets both the tumor cells and their surrounding microenvironment. Pamrevlumab is a fully human monoclonal antibody that inhibits connective tissue growth factor (CTGF), thereby reducing fibrosis and potentially enhancing chemosensitivity. Gemcitabine is a nucleoside analog that inhibits DNA synthesis, acting as a cytotoxic agent directly against cancer cells. Erlotinib is a small molecule inhibitor of the epidermal growth factor receptor (EGFR) tyrosine kinase, interfering with oncogenic signaling and tumor cell proliferation. Clinical studies have examined this combination in locally advanced and metastatic pancreatic cancer, showing promise in improving resection rates and tumor response, especially due to pamrevlumab’s modulation of the tumor stroma[3].
