Pamrevlumab is a fully human monoclonal antibody that targets connective tissue growth factor (CTGF), a key mediator of fibrosis and tumor progression. By binding to CTGF, pamrevlumab inhibits its activity and prevents downstream signaling involved in fibrotic and neoplastic processes[1][2][3]. In oncology settings such as pancreatic cancer, it is being investigated in combination with standard chemotherapy regimens including gemcitabine plus nab-paclitaxel or FOLFIRINOX. Gemcitabine is a nucleoside analog used as chemotherapy; nab-paclitaxel (Abraxane) is an albumin-bound formulation of paclitaxel; FOLFIRINOX combines folinic acid (leucovorin), fluorouracil, irinotecan, and oxaliplatin. The combination aims to enhance antitumor efficacy by targeting both the tumor microenvironment via anti-fibrotic action and directly attacking cancer cells with cytotoxic agents[6].
