PAN004 is an mRNA-based therapeutic developed by Pantherna Therapeutics for the prevention and early treatment of pulmonary edema in early Acute Respiratory Distress Syndrome (ARDS). The drug consists of a cationic lipid nanoparticle (LNP) formulation delivering mRNA encoding a COMP-Ang1 fusion protein, which acts as a Tie2 receptor agonist. By activating the Tie2 pathway in vascular endothelial cells, it aims to stabilize the endothelium, reduce vascular permeability, and prevent pulmonary vascular leakage associated with ARDS. Preclinical studies have demonstrated targeted delivery to lung endothelium, expression and secretion of COMP-Ang1, activation of Tie2 signaling, and pharmacological efficacy in animal models. The product is administered intravenously and is Pantherna’s lead development candidate[1][5].
