**Panadol Osteo** is a modified-release, bi-layer caplet formulation containing **665 mg paracetamol** per tablet, designed for prolonged pain relief. It combines an immediate-release layer for rapid onset and a sustained-release layer using hypromellose polymer for diffusion and erosion-controlled release, maintaining therapeutic plasma levels up to 8 hours. Primarily indicated for persistent pain in **osteoarthritis** and muscle aches like backache, it also provides temporary relief for headache, tension headache, period pain, toothache, colds, flu, and reduces fever. Paracetamol acts centrally to inhibit pain signal transmission to the brain and reduce fever, with bioequivalence to standard immediate-release paracetamol at steady state but lower peak-trough fluctuations for consistent analgesia. Available over-the-counter in Australia and New Zealand as a pharmacist-only medicine (S3), dosed as 2 caplets three times daily (max 6/24h) for adults/children ≥12 years, swallowed whole orally.[1][2][5][9]
