PanChol is a single-dose, live-attenuated oral cholera vaccine (OCV) derived from a seventh-pandemic *Vibrio cholerae* O1 strain. It is genetically engineered to co-express both Inaba and Ogawa antigens and to over-express the non-toxic cholera toxin B subunit (CTB). Additionally, the vaccine targets the toxin-coregulated pilus (TCP), a critical factor for intestinal colonization. PanChol is designed to minimize reactogenicity and be incapable of toxigenic reversion, addressing the limitations of current killed whole-cell vaccines that require multiple doses and show reduced efficacy in children. Clinical data from Phase 1 trials indicate that PanChol is safe, well-tolerated, and highly immunogenic, inducing 100% vibriocidal seroconversion across a wide dose range in healthy adults.
