Panepoxydone is a natural epoxycyclohexenone compound originally isolated from the basidiomycete fungus *Lentinus crinitus* (and also found in *Panus rudis*). It acts as an inhibitor of the NF-κB signaling pathway by preventing the phosphorylation and subsequent degradation of IκBα, thereby blocking the nuclear translocation of NF-κB. Additionally, panepoxydone has been shown to down-regulate the oncogenic transcription factor FOXM1, leading to a reversal of the epithelial-to-mesenchymal transition (EMT). Preclinical research, primarily led by the Mitchell Cancer Institute at the University of South Alabama, has demonstrated that panepoxydone exerts significant antitumor, anti-proliferative, and pro-apoptotic activity against breast cancer cells, particularly triple-negative breast cancer (TNBC) subtypes. It also exhibits antimicrobial properties and inhibits trypanothione reductase in *Trypanosoma cruzi*.
