PANHPVAX is a next-generation, broad-spectrum prophylactic vaccine candidate targeting human papillomavirus (HPV) infection. Unlike current commercial HPV vaccines that use L1 virus-like particles and provide type-restricted protection, PANHPVAX is based on the highly conserved L2 minor capsid protein of HPV. The vaccine antigen consists of a polytope—a string of L2 neutralization epitopes from eight different mucosal HPV types—inserted into a scaffold protein derived from the archaeon Pyrococcus furiosus (thermo-resistant thioredoxin). This construct is further oligomerized via fusion to an OVX313 domain to enhance immunogenicity by forming heptameric nanoparticles. The design aims to induce cross-neutralizing antibodies against a wide range of cancer-causing and some cutaneous HPVs, potentially offering broader protection than existing vaccines. Developed at the German Cancer Research Center (DKFZ), it is currently in early-phase clinical trials as a prophylactic vaccine for HPV infection[1][3][4][5][6].
