Overview

Panitumumab is a fully human monoclonal antibody that targets the epidermal growth factor receptor (EGFR), also known as HER1 or c-erbB-1. It binds specifically to the extracellular domain of EGFR on both normal and tumor cells, competitively inhibiting ligand binding and preventing receptor activation. This blockade inhibits downstream signaling pathways involved in cell proliferation, survival, motility, and angiogenesis. The result is inhibition of tumor cell growth, induction of apoptosis, reduced production of pro-inflammatory cytokines and vascular growth factors, and internalization of EGFR[2][3][5][6]. Panitumumab is primarily indicated for the treatment of EGFR-expressing metastatic colorectal cancer that has progressed following standard chemotherapy regimens[1][7]. It was originally developed by Abgenix using XenoMouse technology and later acquired by Amgen[5].
