Overview

Panitumumab + bortezomib is an investigational combination therapy studied primarily in advanced colorectal cancer and other malignancies. Panitumumab is a fully human monoclonal antibody that targets the epidermal growth factor receptor (EGFR), inhibiting downstream signaling pathways involved in cell proliferation and survival. Bortezomib is a small molecule proteasome inhibitor that blocks the 26S proteasome, leading to accumulation of proteins within cancer cells, disruption of cellular homeostasis, and induction of apoptosis. The combination aims to leverage complementary mechanisms—panitumumab’s inhibition of EGFR-driven tumor growth and bortezomib’s promotion of cancer cell death via proteasome inhibition—to enhance anti-tumor efficacy[1][5]. Panitumumab is developed by Amgen; bortezomib was originally developed by Millennium Pharmaceuticals (now Takeda) and marketed as Velcade or Boruzu[3][4].
