Overview

Panitumumab + chemotherapy refers to the combination of panitumumab, a fully human monoclonal antibody targeting the epidermal growth factor receptor (EGFR), with various cytotoxic chemotherapy regimens used primarily in metastatic colorectal cancer (mCRC) and, in investigational settings, other cancers. Panitumumab binds to EGFR, preventing ligand-induced activation, blocking downstream signaling, and thereby inhibiting tumor cell proliferation, survival, and angiogenesis. Chemotherapy agents commonly used in combination include oxaliplatin-based (e.g., FOLFOX) and irinotecan-based regimens in colorectal cancer, with additional combinations explored in breast, gastric, and head and neck cancers. The combination is generally approved for RAS-wild-type mCRC and is being studied in other malignancies.
