Overview

Panitumumab + irinotecan is a combination therapy used primarily in the treatment of metastatic colorectal cancer. Panitumumab is a fully human IgG2 monoclonal antibody that targets the epidermal growth factor receptor (EGFR), inhibiting its activation and downstream signaling pathways involved in cell proliferation and survival. Irinotecan is a small molecule chemotherapeutic agent that inhibits DNA topoisomerase 1, leading to DNA damage and apoptosis in rapidly dividing cells. This combination has been studied for patients with advanced colorectal cancer who have progressed after standard chemotherapy regimens containing fluoropyrimidines and oxaliplatin. The addition of panitumumab to irinotecan has shown improved outcomes compared to irinotecan alone in certain patient populations, particularly those with wild-type RAS genes[1][3][5][6].
