This is a combination regimen consisting of panitumumab, mitomycin C, and fluorouracil administered with concurrent radiation therapy. Panitumumab is a fully human monoclonal antibody that targets the epidermal growth factor receptor (EGFR), inhibiting its signaling and thereby blocking tumor cell proliferation. Mitomycin C is an antineoplastic antibiotic that acts as an alkylating agent, causing DNA cross-linking and inhibition of DNA synthesis. Fluorouracil (5-FU) is a pyrimidine analog that inhibits thymidylate synthase, disrupting DNA synthesis in rapidly dividing cells. Radiation therapy damages cellular DNA directly through ionizing radiation. This combination has been studied primarily for the treatment of localized squamous cell carcinoma of the anal canal (SCCAC). The addition of panitumumab to standard chemoradiation with 5-FU and mitomycin C increases toxicity without improving outcomes compared to chemoradiation alone[1][2][6].
