Panitumumab + simvastatin is an investigational combination of two drugs, panitumumab and simvastatin, studied primarily in the context of metastatic colorectal cancer with KRAS mutations. Panitumumab is a fully human IgG2 monoclonal antibody that targets the epidermal growth factor receptor (EGFR), inhibiting ligand-induced receptor activation and downstream signaling pathways involved in cell proliferation and survival. Simvastatin is a small molecule statin that inhibits HMG-CoA reductase, blocking the mevalonate pathway and interfering with post-translational modification (prenylation) of proteins such as KRAS. The rationale for this combination was to use simvastatin to modulate mutant KRAS activity—potentially restoring sensitivity to EGFR inhibition by panitumumab in patients whose tumors harbor activating KRAS mutations. However, clinical studies have shown that this approach did not result in meaningful clinical benefit for patients with mutant KRAS metastatic colorectal cancer[1][2].
