Panitumumab + trametinib is an investigational combination therapy consisting of two targeted agents: panitumumab, a fully human monoclonal antibody that inhibits the epidermal growth factor receptor (EGFR), and trametinib, a small molecule inhibitor of mitogen-activated protein kinase kinases 1 and 2 (MEK1/2). This combination has been studied primarily in metastatic colorectal cancer (mCRC) with wild-type RAS/RAF genes. The rationale for combining these drugs is to overcome resistance to EGFR inhibition by targeting downstream signaling through MEK inhibition. Clinical trials have shown that this regimen can induce tumor shrinkage but is associated with significant toxicity, particularly dermatologic adverse events such as acneiform rash. The combination has not demonstrated clear superiority over single-agent anti-EGFR therapy in overcoming acquired resistance and its tolerability limits its clinical utility[1][5].
