Overview

Panitumumab + vemurafenib is a combination therapy used primarily in the treatment of BRAF V600E-mutated metastatic colorectal cancer (mCRC). Panitumumab is a fully human monoclonal antibody that targets the epidermal growth factor receptor (EGFR), inhibiting its signaling and thereby reducing tumor cell proliferation. Vemurafenib is an orally available small molecule inhibitor that selectively targets mutated BRAF-serine-threonine kinase, particularly the V600E mutation, blocking downstream MAPK pathway signaling and inducing apoptosis in tumor cells. The rationale for combining these agents stems from evidence that EGFR reactivation can attenuate the efficacy of RAF inhibitors in mCRC; thus, dual inhibition may overcome resistance mechanisms and improve clinical outcomes. This combination has shown tolerability and disease control in pretreated patients with BRAF-mutant mCRC[1][2][5].
