Overview

Panobinostat + gemcitabine is an investigational combination therapy consisting of two small molecule anticancer agents. Panobinostat is a non-selective histone deacetylase (HDAC) inhibitor that modulates gene expression and induces apoptosis in malignant cells by inhibiting multiple HDAC enzymes, affecting DNA replication, repair, and cell cycle progression. It is approved for use in multiple myeloma in combination with other agents[4][6]. Gemcitabine is a nucleoside analog and metabolic inhibitor that interferes with DNA synthesis by replacing cytidine during DNA elongation, leading to tumor growth arrest and apoptosis. It is approved for various cancers including ovarian cancer, non-small cell lung carcinoma, metastatic breast cancer, and pancreatic cancer[1]. The combination has been studied in early-phase clinical trials for advanced solid tumors; dosing was limited by myelosuppression as the primary toxicity[2].
