Drug intelligence / Profile preview
Pantetheine is a naturally occurring cysteamine amide analog of pantothenic acid (vitamin B5) and an intermediate in the biosynthesis of coenzyme A (CoA). It is the monomeric precursor to pantethine, a widely used dietary supplement. Pantetheine can be phosphorylated by pantothenate kinases (PANK) to form 4'-phosphopantetheine, bypassing upstream rate-limiting steps of the canonical CoA biosynthetic pathway. In preclinical models of progressive multiple sclerosis (MS), pantetheine has been shown to rescue astrocytes from ferroptosis induced by liver kinase B1 (LKB1) deficiency, restoring metabolic resilience and iron homeostasis. It is being investigated by researchers at the University of Illinois at Chicago and the Department of Veterans Affairs as a potential therapeutic strategy for neurodegenerative and demyelinating diseases.
