Overview

A combination therapy consisting of the anthracycline chemotherapy agent doxorubicin hydrochloride and the proton pump inhibitor pantoprazole. This regimen was investigated in a Phase I dose-finding study led by the University Health Network and Princess Margaret Cancer Centre for patients with advanced solid tumors. Doxorubicin hydrochloride works by intercalating into DNA and inhibiting topoisomerase II, thereby preventing DNA replication and transcription. Pantoprazole, traditionally used to reduce gastric acid, was combined with doxorubicin to potentially enhance its antitumor activity by improving drug distribution within the acidic microenvironment of solid tumors and inhibiting autophagy, a mechanism associated with chemotherapy resistance.
