Panulisib (P7170) is an orally bioavailable small molecule inhibitor targeting multiple kinases, including phosphoinositide 3-kinase (PI3K), mammalian target of rapamycin complexes 1 and 2 (mTORC1/mTORC2), activin receptor-like kinase 1 (ALK-1), and DNA-dependent protein kinase. It was developed for its potential anticancer activity, particularly in solid tumors such as estrogen receptor-positive (ER+) breast cancer and non-small cell lung cancer (NSCLC). Panulisib inhibits mTOR at low concentrations and PI3K at higher concentrations, leading to suppression of tumor cell proliferation and induction of apoptosis in some models. The drug has demonstrated potent antitumor effects in preclinical studies but clinical development was suspended during phase I trials due to sponsor decision[1][2][3][4][5].
