Overview

PANVAC + docetaxel is a combination therapy consisting of the poxviral-based cancer vaccine PANVAC and the chemotherapy agent docetaxel. PANVAC is an immunotherapeutic vaccine platform that uses recombinant vaccinia (PANVAC-V) and fowlpox (PANVAC-F) viruses engineered to express tumor-associated antigens carcinoembryonic antigen (CEA) and mucin 1 (MUC-1), as well as three T-cell costimulatory molecules (B7.1, ICAM-1, LFA-3; collectively known as TRICOM). The goal is to stimulate a targeted immune response against cancer cells expressing these antigens. Docetaxel is a taxane-class chemotherapeutic that inhibits microtubule depolymerization, leading to cell cycle arrest and apoptosis in dividing cells. Preclinical data suggest that combining these agents may enhance tumor cell susceptibility to immune-mediated killing by modifying tumor phenotype with chemotherapy while simultaneously stimulating anti-tumor immunity with the vaccine. This combination has been investigated in metastatic breast cancer patients in phase 2 clinical trials for potential improvement of progression-free survival compared to standard chemotherapy alone[1][3][4].
