Overview

PAOPA is a small molecule **allosteric modulator** of the **dopamine D2 receptor**. It is a potent analog of prolyl-leucyl-glycinamide (PLG) and has shown preclinical therapeutic potential for dopaminergic-related illnesses, including animal models of schizophrenia, Parkinson’s disease, and movement disorders such as tardive dyskinesia. PAOPA acts at a distinct allosteric binding site on the D2 receptor, modulating receptor activity and downstream signaling pathways, with demonstrated efficacy in reversing both behavioral and biochemical abnormalities in animal models relevant to schizophrenia and other neuropsychiatric disorders[1][3][4][5][6][7].
