Papain is a cysteine protease enzyme derived from the latex of the papaya fruit (Carica papaya). It functions as a proteolytic enzyme that breaks down proteins into peptides and amino acids. Traditionally used for its anti-inflammatory properties and as a digestive aid in folk medicine and supplements[2][3][4], it has also been applied topically for wound debridement to remove dead or contaminated tissue in acute and chronic lesions such as diabetic ulcers and pressure sores[5][7]. In manufacturing settings it is used in food processing (e.g., meat tenderizer), cosmetics formulation[4], contact lens cleaners[4], and beer clarification. Its mechanism of action involves cleaving peptide bonds via its cysteine protease activity. While widely available as an over-the-counter supplement or ingredient in topical products under various brand names[7], topical drug products containing papain have faced regulatory restrictions due to safety concerns including hypersensitivity reactions[5][7].
