*Parabacteroides goldsteinii AM58-2XD* is a **gram-negative, obligately anaerobic, non-spore-forming, and non-motile bacterium**. It is an antibiotic-resistant gut commensal strain, recently identified as a **probiotic candidate for the treatment of refractory ulcerative colitis (UC)** and possibly other inflammatory gut diseases. Mechanistically, *P. goldsteinii* AM58-2XD alleviates colitis in preclinical models via **colonization of the gut and metabolic enhancement of the valine-isobutyrate (IBN) pathway**. The strain **produces isobutyrate from valine**, which acts as a bioactive metabolite promoting repair of the intestinal mucosa through **activation of the peroxisome proliferator-activated receptor γ (PPARγ) signaling pathway** in intestinal epithelial cells, leading to increased expression of tight junction proteins and improved epithelial barrier function. Genomic analysis revealed multiple antibiotic resistance determinants, enabling colonization during and after broad-spectrum antibiotic administration. The strain is currently at the preclinical research stage and has not been approved for pharmaceutical use[1][3].
