Overview

Paracetamol + morphine + tramadol is a multimodal analgesic combination regimen being evaluated for the management of pain during vaso-occlusive crises (VOC) in adult patients with sickle cell disease. This combination combines paracetamol (acetaminophen), a non-opioid analgesic and antipyretic, with morphine, a strong mu-opioid receptor agonist, and tramadol, a weak mu-opioid agonist and monoamine reuptake inhibitor. By combining these agents with different mechanisms of action, the regimen aims to optimize pain control and reduce overall morphine consumption, thereby minimizing dose-dependent opioid side effects such as respiratory depression, sedation, and constipation.
