paracetamol + glycerol

paracetamol + glycerol is a combination adjuvant therapy consisting of oral glycerol and high-dose rectal paracetamol (acetaminophen). It was evaluated by the Kamuzu University of Health Sciences (formerly the University of Malawi College of Medicine) as a potential treatment to improve outcomes in children with bacterial meningitis. Glycerol serves as an osmotic agent intended to reduce intracranial pressure and cerebral edema, while paracetamol acts as an antipyretic and anti-inflammatory agent to mitigate fever and cytokine-mediated damage. A Phase 3, randomized, double-blind, 2x2 factorial trial conducted in Malawi demonstrated that this combination, when added to standard antibiotic treatment (such as ceftriaxone), did not significantly improve mortality or morbidity rates in pediatric patients.
