Drug intelligence / Profile preview
Paracetamol + morphine + ketamine is a multimodal analgesic combination therapy being evaluated for the management of pain during vaso-occlusive crises (VOC) in adult patients with sickle cell disease (SCD). The combination utilizes three distinct mechanisms of pain relief: paracetamol (acetaminophen) acts primarily through central analgesic pathways, morphine is a strong opioid agonist targeting mu-opioid receptors, and ketamine is an NMDA receptor antagonist that acts as a co-analgesic to prevent opioid-induced hyperalgesia and reduce overall opioid consumption. This combination is being studied in the PAMAVOC adaptive platform trial sponsored by Assistance Publique - Hôpitaux de Paris (AP-HP) to optimize pain management protocols and minimize opioid-related adverse effects.
