Nuberol Forte is a fixed-dose combination of paracetamol (acetaminophen) and orphenadrine citrate, primarily used as an analgesic and skeletal muscle relaxant. Paracetamol provides antipyretic and analgesic effects, likely through the inhibition of cyclooxygenase enzymes and subsequent reduction in prostaglandin synthesis within the central nervous system. Orphenadrine acts as a centrally acting muscle relaxant, exhibiting anticholinergic and antihistaminic properties; its muscle relaxant effects are attributed to its activity in the brain stem, where it antagonizes NMDA receptors. This combination is frequently prescribed for the symptomatic relief of painful musculoskeletal conditions, including chronic neck pain, low back pain, and muscle spasms. It is manufactured and marketed by The Searle Company, particularly in the Pakistani pharmaceutical market.
