Drug intelligence / Profile preview
Parathyroid hormone (recombinant human parathyroid hormone 1-84) is a recombinant version of the endogenous 84-amino acid human parathyroid hormone (PTH). It acts as an agonist at the parathyroid hormone 1 receptor (PTH1R) and parathyroid hormone 2 receptor (PTH2R), regulating calcium and phosphate homeostasis, renal calcium reabsorption, active vitamin D synthesis, and bone turnover. Originally developed by Allelix Biopharmaceuticals and NPS Pharmaceuticals, it was approved under the brand name Preotact (in Europe) for postmenopausal osteoporosis and later under the brand name Natpara/Natpar (by Shire and subsequently Takeda) as a replacement therapy for chronic hypoparathyroidism. However, due to persistent manufacturing and supply issues, including protein and rubber particle formation, Takeda permanently discontinued global manufacturing of the drug at the end of 2024.
