Drug intelligence / Profile preview
Parathyroid hormone-related peptide (PTHrP) is an endogenous proteinaceous hormone that belongs to the parathyroid hormone family. It is widely expressed in various tissues and plays critical autocrine, paracrine, and intracrine roles in regulating bone development, tooth eruption, smooth muscle tone, and transepithelial calcium transport. Structurally, its N-terminal region shares high homology with parathyroid hormone (PTH), allowing it to bind and activate the parathyroid hormone 1 receptor (PTH1R). While primarily an endogenous signaling molecule, synthetic fragments such as PTHrP(1-36) have been investigated in clinical trials as potential anabolic therapies for osteoporosis. Additionally, PTHrP is clinically significant as a biomarker, as its hypersecretion by certain tumors is the primary driver of humoral hypercalcemia of malignancy (HHM).
