Overview

The **parent ADC** is a non-pH-sensitive antibody-drug conjugate (ADC) targeting the hepatocyte growth factor receptor (cMET), utilized by Mythic Therapeutics as a comparative control for their lead candidate, MYTX-011. This molecule consists of an anti-cMET monoclonal antibody conjugated to the microtubule-disrupting payload monomethyl auristatin E (MMAE) via a protease-cleavable valine-citrulline (vc) linker. Unlike MYTX-011, which is engineered for pH-dependent dissociation to enhance payload delivery in tumors with varying cMET expression levels, the parent ADC maintains standard binding kinetics. In preclinical studies, it has been used to demonstrate that pH-sensitive engineering significantly increases the delivery of free MMAE and enhances anti-tumor activity in xenograft models of non-small cell lung cancer (NSCLC), gastric, and head and neck cancers.
