Paridiprubart is a fully humanized IgG1 kappa monoclonal antibody that targets and inhibits toll-like receptor 4 (TLR4), a key innate immune receptor involved in mediating inflammation and fibrosis. By binding with high affinity to TLR4, paridiprubart blocks its dimerization and activation, thereby suppressing downstream inflammatory signaling pathways such as NFκB, AP-1, and IRF3. This mechanism reduces cytokine release (including TNFα, IL-6, CXCL10, IFNβ) and physiological responses associated with acute inflammatory conditions. Paridiprubart is being developed by Edesa Biotech for both acute and chronic diseases characterized by dysregulated innate immunity—most notably for acute respiratory distress syndrome (ARDS), including COVID‑19-induced ARDS, as well as progressive lung fibrosis. Clinical studies have shown promising results in reducing mortality among critically ill patients with ARDS[1][2][5][6][7].
