Overview

Parkin activators are a class of small molecule allosteric modulators being developed by Nine Square Therapeutics for the treatment of Parkinson's disease and metabolic disorders. These compounds act as molecular glues that enhance the interaction between phospho-ubiquitin (pUb) and the E3 ubiquitin ligase Parkin (PRKN). By promoting Parkin's activity, these activators facilitate mitophagy—the selective degradation of damaged mitochondria—thereby restoring cellular mitochondrial function and providing neuroprotection to dopaminergic neurons. The program is currently in preclinical development and has received significant research funding from the Michael J. Fox Foundation.
