Overview

Paroxetine + thyroid hormone refers to the combination of a selective serotonin reuptake inhibitor (SSRI), paroxetine, with a thyroid hormone (most commonly triiodothyronine/T3 or levothyroxine/T4). Paroxetine is an antidepressant that works by inhibiting the reuptake of serotonin in the brain, thereby increasing its availability and improving mood. Thyroid hormones regulate metabolism and have been shown to modulate neurotransmission, particularly serotonergic pathways. The rationale for combining these agents is based on evidence that thyroid hormones may augment or accelerate antidepressant response in some patients with major depressive disorder, especially those who are refractory to standard treatments. However, clinical studies have not consistently demonstrated significant benefit from adding T3 to paroxetine in nonrefractory depression; moreover, this combination may increase adverse effects[1][4][6]. There is also evidence suggesting potential drug–drug interactions between SSRIs like paroxetine and levothyroxine that can reduce the efficacy of thyroid replacement therapy and potentially induce hypothyroidism[5].
