Overview

parp4 siRNA nanoparticles are preclinical-stage therapeutic candidates developed by researchers at the University of Texas Health Science Center at San Antonio. These nanoparticles consist of small interfering RNA (siRNA) designed to silence poly(ADP-ribose) polymerase 4 (PARP4), chemically conjugated and encapsulated within poly(lactic-co-glycolic acid) (PLGA) nanoparticles. PARP4 acts as a co-regulator of the estrogen receptor (ER), and its overexpression is associated with hormone-dependent breast cancer progression, metastasis, and resistance to endocrine therapies. By knocking down PARP4 expression, these nanoparticles aim to inhibit cell cycle progression, reduce local aromatase activity, suppress metastatic potential, and resensitize hormone-resistant breast cancer cells to endocrine therapies.
