Parsaclisib + itacitinib is an investigational combination therapy composed of two small molecule inhibitors developed for the treatment of hematologic malignancies and solid tumors. Parsaclisib is a selective inhibitor of phosphatidylinositol 3-kinase delta (PI3Kδ), which plays a key role in the growth and survival of B-cell cancer cells. Itacitinib is a selective Janus kinase 1 (JAK1) inhibitor, targeting cytokine signaling pathways involved in immune cell regulation and inflammation. The combination aims to modulate both tumor cell survival and the tumor microenvironment by inhibiting these complementary pathways, potentially enhancing antitumor immune responses. Both agents are being developed by Incyte, with clinical studies evaluating their safety, tolerability, pharmacodynamics, and preliminary efficacy in patients with advanced or metastatic solid tumors as well as B-cell malignancies[2][4][8].
