Parsaclisib + rituximab is an investigational combination therapy used in clinical trials for relapsed or refractory B-cell lymphomas[1][3][5]. **Parsaclisib** is a potent, highly selective phosphoinositide 3-kinase delta (PI3Kδ) inhibitor; it blocks intracellular PI3Kδ signaling, which is critical for B-cell proliferation and survival[3]. **Rituximab** is a chimeric anti-CD20 monoclonal antibody that depletes B cells by targeting the CD20 antigen on their surface, leading to antibody-dependent cellular cytotoxicity, complement-dependent cytotoxicity, and direct apoptosis[8]. The combination aims to improve efficacy in B-cell malignancies by synergistically targeting malignant cells through complementary mechanisms—one blocking survival signaling, the other inducing depletion. The combination has been evaluated in phase 1 studies (CITADEL-112) for relapsed/refractory B-cell lymphoma, showing promising efficacy and a manageable safety profile[1][3][5].
