Overview

Parsaclisib + ibrutinib is a combination of two small molecule kinase inhibitors used in the treatment of B-cell malignancies. Parsaclisib is a potent and highly selective inhibitor of phosphoinositide 3-kinase delta (PI3Kδ), which plays a key role in B-cell receptor signaling and survival. Ibrutinib is an irreversible inhibitor of Bruton's tyrosine kinase (BTK), another critical enzyme in the B-cell receptor pathway. By targeting both PI3Kδ and BTK, this combination aims to disrupt multiple signaling pathways essential for malignant B-cell proliferation and survival. Both drugs have shown clinical benefit as monotherapies or in combination with other agents for relapsed/refractory mantle cell lymphoma (MCL), marginal zone lymphoma (MZL), chronic lymphocytic leukemia (CLL), Waldenström’s macroglobulinemia (WM), and chronic graft versus host disease (cGVHD). The combination has been explored clinically for enhanced efficacy in relapsed/refractory B-cell lymphomas[2][4][8].
