Overview

Partially HLA-matched unrelated donor cells refer to a cellular therapy product used in the microtransplantation (MST) protocol developed for patients with acute myeloid leukemia (AML) who are ineligible for standard allogeneic stem cell transplantation. This approach, specifically studied by the University of Sydney in the SCRAMBLE trial, involves the infusion of donor peripheral blood stem cells or marrow cells following consolidation chemotherapy. Unlike traditional transplantation, MST does not require post-infusion immunosuppression, aiming to induce a transient micro-chimerism that triggers a graft-versus-leukemia (GVL) effect to eradicate residual disease while minimizing the risk of graft-versus-host disease (GVHD).
