PAS-002 is an investigational DNA-based tolerizing vaccine being developed by Pasithea Therapeutics for multiple sclerosis, designed to induce immune tolerance to the myelin-associated protein GlialCAM rather than stimulate an immune response. Preclinical studies in relapsing-remitting experimental autoimmune encephalomyelitis mouse models showed that intramuscular administration of PAS-002, a proprietary plasmid encoding GlialCAM, delayed disease onset, reduced peak disease severity, decreased relapse severity, and prevented disease in approximately half of treated animals, supporting its potential as a prophylactic “inverse vaccine” to prevent autoimmune demyelination by dampening pathogenic responses driven by molecular mimicry between Epstein–Barr virus antigens and GlialCAM.[1][2][3][11]
