Overview

PAS-nomacopan is a long-acting recombinant protein therapeutic designed as a bispecific inhibitor of complement C5 and leukotriene B4 (LTB4), developed for intravitreal injection in the treatment of geographic atrophy (GA) secondary to dry age-related macular degeneration. The drug leverages PASylation technology to extend its half-life in the eye, potentially allowing for dosing intervals of three months or longer—meaning only three or four injections per year may be needed. By inhibiting both complement C5 and LTB4 pathways, PAS-nomacopan aims to slow GA lesion progression while reducing the risk of choroidal neovascularization (CNV), a complication associated with current complement-only inhibitors. The drug is being developed by Akari Therapeutics and has completed preclinical development with plans for an IND submission in 2025[1][2][3][5][6].
