Overview

Pasireotide + everolimus is a combination therapy of two pharmaceutical agents used primarily in the treatment of advanced neuroendocrine tumors (NETs), including pancreatic NETs, and has also been studied in other cancers such as hepatocellular carcinoma and medullary thyroid carcinoma. Pasireotide is a second-generation somatostatin analogue that binds to multiple somatostatin receptor subtypes, inhibiting hormone secretion and cell proliferation. Everolimus is an mTOR inhibitor that blocks the mammalian target of rapamycin pathway, leading to reduced cell growth, proliferation, angiogenesis, and glucose uptake. The combination aims for synergistic antiproliferative effects by dual inhibition of insulin-like growth factor 1 (IGF-1) signaling (via pasireotide) and mTOR signaling (via everolimus). Both drugs are developed by Novartis[2][3][1][8].
