Pasireotide LAR is a long-acting formulation of pasireotide, a synthetic cyclic hexapeptide and somatostatin analog. It is administered as an intramuscular injection once every four weeks. Pasireotide acts by binding to multiple somatostatin receptor subtypes (with high affinity for types 1, 3, and 5), leading to inhibition of adrenocorticotropic hormone (ACTH) secretion from the pituitary gland and subsequent reduction in cortisol production. This mechanism makes it effective for treating Cushing’s disease in adults who are not candidates for surgery or have failed surgery. Pasireotide LAR is also indicated for acromegaly in patients who cannot be treated with surgery or did not respond to surgical intervention[2][4][6][8]. The drug was originally developed by Novartis and is marketed under the brand name Signifor LAR.
