Pasodacigib (BAY 2862789) is an orally bioavailable, small molecule inhibitor of diacylglycerol kinase alpha (DGKα). Developed as a novel immuno-oncology agent, it selectively inhibits DGKα—an enzyme that regulates T-cell receptor signaling and is implicated in cancer cell survival and proliferation. By blocking DGKα activity, pasodacigib enhances T-cell activation and effector function by increasing diacylglycerol levels within T-cells. This leads to improved anti-tumor immune responses through increased T-cell proliferation, cytokine production, and cytotoxicity. Additionally, inhibition of DGKα may directly induce apoptosis in tumor cells that overexpress this enzyme. Pasodacigib is being investigated primarily for the treatment of advanced solid tumors—including non-small cell lung cancer—in Phase 1 clinical trials[5][3][4].
