Drug intelligence / Profile preview
PASylated anti-CD98hc Anticalin (also known as D11vs-PAS200) is a preclinical-stage, half-life-extended Anticalin protein designed for tumor imaging and potentially theranostic applications. Developed by XL-protein in collaboration with academic partners at the Technical University of Munich, the agent is based on the human lipocalin 2 (Lcn2) scaffold engineered to bind the extracellular domain of human CD98 heavy chain (CD98hc, encoded by SLC3A2) with picomolar affinity. CD98hc is a type-II transmembrane glycoprotein overexpressed in various solid and hematological malignancies, where it acts as a chaperone for amino acid transporters (like LAT1) and regulates integrin signaling. To optimize its pharmacokinetic profile for in vivo imaging, the Anticalin is genetically fused to a PAS sequence (PAS200, a structurally disordered polypeptide of proline, alanine, and serine), which increases its hydrodynamic volume and retards renal clearance. When conjugated with deferoxamine (Dfo) and radiolabeled with zirconium-89 (89Zr), the tracer enables high-contrast positron emission tomography (PET) imaging of CD98hc-positive tumors, demonstrating strong tumor accumulation and favorable tumor-to-blood ratios in preclinical models.
